The low molecular weight glutenin subunit (LMW-GS) is the main component of wheat gluten. The LMW-GS is one of the main factors that determine the nutritional and technological properties. Because of their large number and considerable heterogeneity, they are difficult to be separated by Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE). The studies about LMW-GS are rare compared with HMW-GS and focus on primitive wheat and durum wheat.this paper study the LMW-GSs gene utilizing the common wheat cultivars: PH 82-2-2. the results as fellow:1. We designed three pairs of primes according to published primitive wheat LMW-GS gene sequences and obtained nine DNA fragment from 700 to 1500. They are target sequences.Through analysis and nucleotide Blast of those sequences and protein, N-terminal and C-terminal sequences are conserved. The difference of them is single resides insection or detection. The main repetitive unit of repetitive sequences is PFSQQQ. The repeats number is from 1 to 11. Otherwise, there have another repeats, such as QPVLPQ,PILPQQ,PIIIL. The numbers of2. repeats is from 2 to 3. Differences of numbers and sequence of repeats are responsible for some variation in amino sequence of LMW-GSs. To classify these LMW-GS groups based on the function aspects of dough, we divided the nine LMW-GS into six types based on the position of the cysteines. According above, we obtained the common structural model of LMW-GS in common wheat.3. We can predict the location of these genes based on amine acid sequence similarity to previously reported sequence of loci, which have been identified.Glu-D3G5,G1,G2 and G6 encoded by Glu-D3。G8,G7 and G3 encoded by Glu-A3。G4 and G9 encoded by Glu-B3。...
