Lily(Lilium, spp) belongs to Lilium, Liliaceae. It is very famous ornamental plants and widely used in food and pharmacy industry as well. But due to the low propagation rate of lily bulblet and its susceptivity to virus invasion, the product quality of lily remains very poor. Quick propagation technique can produce large number of plantlets rapidly; Polyploid plants by Chromosome doubling induction have such good characteristics like strong plant, thickening scale etc. For these reason, it is widely accepted that studies on quick propagation and chromosome doubling of Lily is important and significant both in theoretic aspect and in economic aspect. The thesis focus on two main parts, one is the culture medium in quick propagation of LIUYANG lily; The other is chromosome doubling of the same plant.The main conclusions of our studied are listed below:(1) The ingredients and combination of hormone has a significant effect on the induction of lily bulblet and shoot proliferation lily. In the shoot induction, BA plays a very important role; Dark culture and thicken scale have positive effect on bulblet induction. The shoots proliferation has individual variety. Generally speaking, root growing is easy and 1/2 MS is best suitable culture medium. It also found that AC(active carbon) is beneficial to root growing. The best suitable induction medium of LiuYang lily is MS+BA2.0 mg/L+NAA0.2 mg /L; The best proliferation medium is MS+BA2.0 mg/L+NAA0.2 mg/L; The best growing root medium is 1/2MS+IBA0.8+AC.(2) In the scope of this study, the best concentration of colchicine is 0.1% in chromosome doubling induction of lily, under the concentration, the optimize time is 48h, and shoot rate is 96%, the rate of abnormal growing phenomenon is 32%.(3) In the scope of this study, the best concentration of Oryzalin is 0.1%, the optimize time is 3h, and the shoot rate is 100%, the rate of growth abnormal phenomenon is 46.7%.(4) According to the Flow Cytometry (FC) test, both Oryzalin and colchicine induced polyploid lily successfully, and the doubling rate is higher when treated lily with Oryzalin.(5) In this paper, Oryzalin used firstly in chromosome doubling in China, and the result showed that: Oryzalin have many advantages :it is much less toxic, it needs shorter treat time,the shoot rate and the doubling rate are higher, and the apparent induction rate is higher from morphologic reservation. So we are confident that Oryzalin can be a promising alternation of colchicine in chromosome doubling.
