Study On Chromosome Doubling Of Polyploidy Lilium Lancifolium In Tissue Culture

In our experiment, we took the Lilium lancifolium as experimental material.. In order to obtain the polyploidy Lilium lancifolium which is stronger and has higher ornamental value, the tissue culture and several methods of chromosome doubling were studied deeply in our experiment. The results were follows:(1) Studied on the 0.1%HgCL2 as sterilization agent, the best sterilization time is 13 min, and it was more hurt to bacteria and less hurt to plant.(2) Studied on the flakes of bulbus as explant, to produce bud points, the best medium was MS+2.0mg/L6-BA+0.2mg/LNAA.(3) Studied on the flakes of bulbiet as explant, to produce bud points, the best medium was MS+1.0mg/L6-BA+0.1mg/LNAA.(4) The best medium for rooting was MS+1.0mg/LIBA(5) In the test of using the medium with colchicines to treat flakes, the best Combination was adding colchicines 50mg/L into the medium treated flakes for 14 days, and the polyploidy Mutagenic n rate reached75%(6) In the test of using the medium with colchicines to treat callus, the best Combination was adding colchicines lOmg/L into the medium treated callusfor 21 days, and the polyploidy Mutagenic n rate reached 70%。(7) In the test of soaking the flakes into colchicine, the best Combination was putting the flakes into 0.1%colchicine for 12 hours, and the polyploidy Mutagenic rate reached 50%.(8) In the test of soaking the pre-cultured flakes into colchicine, the best Combination was putting the flakes which was pre-cultured for 14 days into 0.04%colchicine for 9 hours, and the polyploidy Mutagenic rate reached 70%(9) In the test of soaking the cultural plantlets into colchicine, the best Combination was putting the small bulbus into 0.1%colchicine for 24 hours and the polyploid Mutagenic rate reached 57.14%,and mortality rate was 21.4%(10) In polyploids identification,we found hexaploid which has 72 chromosomes by Root tip chromosomes observation.what’s more we also found mixoploid with 60、57、48、44、40、 38、29 chromosomes in Stoma observation,we found the length of Stoma was increased by 46.39%,and the density of Stoma was descended by 70.23%.
