Cotton plant is frightened by lots of pests and diseases during its life, and this leads to the serious loss of yields and lower fiber quality. Among these diseases, Verticillium wilt is a worldwide one and very popular in many countries. In recent years, this disease had spread widely and became more and more serious which became an obstacle to increasing fiber output and fluctuate cotton product. Therefore, "the cancer of cotton" is named after it in China. Moreover, the study of molecular biology of Verticillium wilt in cotton lagged behind other crops and further affects the progress for breeding Verticillium wilt resistant varieties.The research on the identification method, resistant inheritance and SSR molecular markers of Verticillium wilt were carried out by using the intraspecific cross of Upland cotton and the interspecific cross between G.barbadenseXG.hirsutum. The intention was to explore the identification method of Verticillium wilt resistant, understand the characteristics of resistant inheritance of Verticillium wilt, and search the molecular marker linked with the resistant gene. It would build a basis for mastering regulation of Verticillium wilt resistant inheritance and cloning the resistant gene. The main results as follows:1 , The compared studies of identity and difference of field and greenhouse identification of Verticillium wilt resistant indicated that the difference between field and greenhouse. Therefore, it is suggested inheritance identification of Verticillium wilt resistance more accurately should apply greenhouse identification method by using the seedling of f2:3 family.2, Genetic analysis of resistance of Verticillium dahliae by parents, F2 population and F2:3 family of Upland X Upland and G.barbadense X Upland indicated that II 15-3493 (G.barbadense line) and Arcot-! (Upland line) were resistant to Verticillium dahliae Anyang strain, and this resistance were controlled by one dominant major gene with somemodified gene.3, The resistant ability of the resistant variety of G.barbadense, F2 population and F2:3 family of G.barbadense Upland was stronger than that of Upland variety and its offspring. This indicated the resistance from G.barbadense was stronger than that from Upland lines. The resistant mechanism of G.barbadense variety or plants of G.barbadense type in the cross maybe prevent disease infection and extension further while that of Upland type mainly prevented extension.4, 216 pairs SSR primers were amplified among the DNA of P1, P2 , F2 and their segregated bulk of the crosses of G.hisutum G.hirsutum and G.barbadense X G.hirsutum. The results indicated that 19 pairs of SSR primers had polymorphism between parents of cross of Upland X Upland, but no polymorphism between DNA pools. 53 pairs of SSR primers had polymorphism between parents of cross of G.barbadensey. Upland , and four pairs of SSR primers with polymorphism between gene pool. It indicated the inheritance background of G.barbadnse was very different from that of Upland. So the population with higher genetic diversity was better for construct a linkage map.5, The PCR amplification products of F2 individual from the cross . II15-3493 Shi he-zi 875 were analyzed by using MAPMAKER/EXP(VERTION 3.0b) and MAPMAKER/QTL(VERTIONl.lb) soft program. There were three SSR loci related with the genes of G.barbadense Verticillium wilt resistance. They were SKH1- SKH2 and SKH4, covered genetic distance of 88cM. It was assured elementary that Verticillium wilt resistant gene (VRB1) located between SKH2 and SKH4, and linked with SSR locus SKH4 tightly with a distance of 13.1cM, and 28 cM from SKH2 locus..The innovation of this research is that SSR molecular marking technology was introduced to find genes relevant to Verticillium wilt resistance by using the F2 :3 family as the population for resistance identification , which can make the result much objective and accurate . This will build the basis for isolating and cloning the Verticillium resistant genes in future.
