To deeply understand the function of WSSV VP37 on the virus atttachment, VP37 was recovered by the means of electroelution and reversible staining. It was observered that the the binding of WSSV on the primary cultured cell of shrimp Penaeus. chinensis was depressed by VP37 polyclonal antibodies. In addition, the depressing effect of VP37 polyclonal antibodies against WSSV binding to cell membrane component was observered in microplate. Results of experiments demonstrated that polyclonal antibodies of VP37 could partilly depress WSSV binding to cell and cell component. This farther proves that VP37 was attachment protein or one of the components of WSSV attachment proteins. Five series of monoclonal antibodies was compared considering the effect of monoclonal antibodies on blocking WSSV binding to cell,they are 1A4, 6A4, 2B2, 4A1 and 1D5. Two series of monoantibodies were found that can partilly depress the WSSV binding to cell, 2B2 and 1D5, the others showed no effect.Sodium dodecyl sulphate-polyacrylamide gel electrophoresis(SDS PAGE) of WSSV showed that at least 7 structural polypeptides were observered: 252. 1,207.2,201.7,172.8,156.9, 128.8 and 105.7Kd.And the relative amount of the bands was analyzed by software GelproAnalysis,which showed that the relative amount order was 252. 1>105. 7>128. 8>201.7>207. 2>172. 8>156. 9Kd0 Two high molecular weight proteins, with MWs of 159Kd and 233Kd were showed when the virus proteins were blotted onto the NCM and binded with labeled cell membrane proteins. More test need to launch to prove the function of the two proteins when considered the limitation of blotting experiment.
