Six ectomycorrhizal fungi of Betula platyphylla were screened from the eleven fungi which were collected under the mixed forest of Betula platyphylla Suk..They are Agaricus sihaticus Schaeff., 009, 035, 044, GT005 and LH008. Through observing of the ectomycorrhizal exterior forms and tissue sections, we found there were mantle of fungi tissue which enclosed the root, emanating hyphae of the rhizoplane, and the Hartig net which was formed by the hyphae between the epidermal and cortical cells. These characteristics revealed that ectomycorrhizal fungi and Betula platyphylla had found symbiosis relationship.The optimum growth temperature, optimum growth carbon source, optimum growth nitrogen source and optimum growth pH value of six ectomycorrhizal fungi were determined from test of cultural and physiological characteristics. The optimum growth temperature of Agaricus sihaticus, 009, LH008 and GT005 was 30℃, belonging to high temperature fungi, and that of 035 and 044 was 25℃, belong to moderate temperature fungi. The optimum growth pH value of these fungi was 6.5 except that of GT005 was 4.0. The optimum growth carbon source of Agaricus silvaticus, 009 and GT005 was sucrose, that of 035 was glucose, and that of 044 and LH008 was maltose. The optimum growth nitrogen source of these fungi was Asparagine except that of 009 was NaNO3.Through inoculating to Betula platyphylla, we found that these ectomycorrhizal fungi can evidently improve the biomass, content of chlorophyll, photosynthetic rate, content of nitrogen and phosphorus, activity of hydrogen peroxidase, and root vigor of seedling of Betula platyphylla.
