Subcutaneous and perirenal adipocyte plasma membranes of pig were obtained by ultrasonic disruption, mash and vigorous inversion, and assessed by 10%-SDS-PAGE electrophoresis. The results demonstrate that ultrasonic disruption is the best, vigorous inversion is the last, weight molecules of membrane proteins are 15 kd-118 kd.Nine goats were allotted to 3 groups randomly and each group included 3 goats respectively. The I group was immunized with membrane proteins 4.6ml (5mg) ,and boosted 9.4ml (10mg) two times after three weeks and five weeks, the II group was immunized with membrane proteins 7ml (7.5mg) ,and boosted 14ml (15mg) two times after three weeks and five weeks, the III group was immunized with membrane proteins 9.4ml (10mg) , and boosted 18.8ml (20mg) two times after three weeks and five weeks, collected positive antisera and determined the liter. The result indicated that the highest titer is 1:5120.Isolated and purified the highest antisera, determined the specificities. The results showed that antisera against pig adipocyte plasma membrane have species-specific , tissues-specific conspicuously, and site-specific inconspicuously.
