Objective: To establish standardization biological index of KM mice.Method: One hundred and twenty litters of mice aged 0~36w were randomly selected to test the neonatal development parameters, the blood routine indexes , the blood biochemical index, the body length ,the tail length, and the weights of viscera of each KM mice aged 28, 56 and 112d with biochemical analyzer caliper or balance. One hundred and eighty 21-day-old mice (1: 1) were randomly selected and were divided into six group according to feed intake, breeding, and non-breeding. And the life span of the mice were recorded. Forty KM mice were provided by Beijing institute of biological product (Beijing), ChangChun institute of biological product (Changchun), North china pharmaceutical factory (Huabei), institute of laboratory of Chinese Medical Science Academy(CAMS), and N1CPBP in Kunming subcolonies, respectively. 13 loci were determined by biochemical isoenzyme electrophoresis.Result: The average weight of KM mice aged was 36w 48.84g( ± 7.3110), the mean body length of KM mice were 201.50 ( ±6.5970)mm (male) and 204.15 ( ± 5.7700)mm (female) respectively. The first litter rate of mice aged 30d was 94%, the average litter size was 11.9(1.9687) the average survival rate was 88.60%( ± 7.8350), the content of raw fat was 21.03%( ± 7.0709). The mean life span was 582.5 ± 177.73, 730.25 ± 167.39, and 548.75 - 148.66 for normal feeding, restricted feeding, and ad lib feeding mice, respectively. There was no difference between normal feeding and ad lib feeding mice (P>0.05), while the life span of restricted feeding mice was significantly longer than that of normal feeding and ad lib feeding mice (P<0.001).Only b type was found on the allelesloci of Akp1, Es1.and Trf in five KM subcolonies , and only a type was found on loci of Ce2 in Beijing and Huabei Kunming colonies.On the loci of Es3, a type, ac type, and ab type was not found in Kunming colonies besides the Beijing and the Changchun subcolonies . A single type of Hbb(s) , Idh1 type was found in CAMS colonies. Alleles distributing of five KM colonies was in a state of balance at some loci .The result of genetic distance was related to the origin of colonies.Conclusions: (1) The biological properties of KM mice in ChangChun institute of biological product (Changchun) was tested, and it was the first time in twelve cooperational units to test the life span of Kunming mice from it was introduced, which provided dependable data for the standardization of KM mice. (2)The result from the comparation of the genetic biological loci in five Kunming subcolonies show that there are difference in Kunming subcolonies, and the standardization is required.
