Drought is one of the most important non-biological factors diminishing soybean pruduct.The drought resistance is various in different period.During the past decade, a number of genes and upstream regulatory elements responding to the stresses have been isolated and well characterized. In the present study, studying the drought resistant for shoot by BSA and AFLP technology, different display was adopted in a attempt to isolate soybean genes that are regulated by stress, the QTL mapping of the drought resistance are also determined. The main experimental results are as follows:1. Finding out 9 AFLP molecular mark about resistance to drought. The DNA of soybean diffrent fragment were solated and Z1 was characterized. Zl DNA is 312 bp in lengh. Southern blot analysis shows that Z1 gene is present as a single-copy sequence in the soybean genome.2.RT-PCR analysis indicated that Zl transcripts were more abundant in leaf and root but only a little in stem, and Zl gene was not significantly induced by drought and salinity. So the Zl gene DNA sequence is different in the two presents genome.3.Two QTLs were detected for drought resistance of sprouting by composite interval mapping. One QTL nominated CYL1 was detected in the G2 linkage group on the region between satt329 and satt377. The percentage of observed phenotypic variance attributable to CYL1 was 18.9%. The other QTL was detected in the g6 linkage group and between the satt460 arid satt100.
