Mosaic virus is a serious disease of soybean in China,which is caused by the virus.It dramatically reduce soybean yield.So it is urgent to study the disease-resistant mechanism at the integral level and to improve disease resistance in soybean using the modern molecular biology.We have demostrated the inheritance of resistence of 95-5383 by studying the progenies of the crosses of resisant x susceptible. We have used cDNA-AFLP technology to analyse the gene express after the intrusion SMV3, reclaiming the 108 differention fragment related to resistance gene, for ground the after work. The main results are summarized as followings:1. 6 soybean accessions have been identified for the resistance to SMV3 by artificially inoculation with SMV3, a strong virulent strain in China. Symptoms are different in different varieties when inoculated with the same SMV strain, showing there are interaction between soybean varieties and SMV strains.2. One highly resistant soybean line(95-5383)was crossed with four susceptible soybean varieties IA2020.The F1 and F2:3 individuals were identified for SMV resistance by inoculation with SMV3.The results showed that in the crosses of resistant x susceptible, F1 were susceptible and F2:3 populations segregated in 1:2:1 ratio, indicating that 95-5383 caues one recessive gene that confer resistance to SMV.3. Parents and offsprings of Zhe chun 3 were used in the experiment for identification of their resistance to SMV and molecule assistant breeding select. According to Nei-Li similarty coefficients computed by SSR data, a dendrogram with 3 groups was constructed by the UPGMA.It is shown that the cultivars with the same ecotype, the same geographical origin and the approximate pedigree were clustered together. The genotypes within distinct clusters may possess different resistance genes that could be exploid in soybean breeding programs. The result indicating amplified the same fragment to comparison maybe carry the same resistance gene as was comparison,but others did not amplified the same fragment to comparison maybe carry the different resistance gene as was comparison opposition.4. cDNA-AFLP was employed to analyze expression profiles of soybean during artificially inoculation with SMV3 before ten period,though PA electrophoresis find the different fragment comparative enrich 0,4h, 12h, 24h, 48h five period.5. We analysis the gene expression of soybean mosaic virus with cDNA-AFLP.The most suit prime combination are E-CGA M-CG and E-CT M-ATT. Thereinto the 2+2 prime combination amplified the fragments more than the 3+2, and 2+3 more than the 3+3. Namely with the selected the bp increase, the amplificed bands has reduced. It is consistent with the theory and probability, because add 1 bp the fragments will be reduce 1/4.3+3 prime combination bands fewer, information content fewer, it is not suit for identificating the gene expression. So the better of the 2+2,3+2 prime combination.6. Using 36 pairs E1-6 and M1-6 primer combination analysis gene expression fragment, according to inoculate 0,4,12,24,48 hours using cDNA-AFLP, amplified 108 fragments, each time...
