By the techonology of mRNA differential display we analysised the gene expression of nucleus of the wheat . The material we use is T-type wheat cytoplasmic male sterile lines 75-3369A and their maintainers 75-3369B .We choose 6 differential materials, which from 3 differenr times. We use oligodTi2CG as arbitrary primer and 11 random primers for PCR amplification . Most of the fragments are the same between A and B, which showed that the heredity background of the material we use is good. Sixteen positive clones were identified out of the 44 fragments by reverse-northern hybridization . After querying in genebank , four clones were found have no high similarity with other sequences in plants' tissues and twelve clones were found have high similarity with other sequences in which two have high relativity with sterility. They are gene of Triticum aestivum cytosolic starch phosphorylase and actin. The four clones which have no high similarity with other plants' sequences all have ORF, so we may regard them as unknown genes. The result we got, offer condition for isolation the whole-length gene about sterility and illuminate the mechanism of CMS.
