Objective: The purpose of the present work is to transfer the effective element of Radix Panacis Quinquefolii into carrot by somatic hybridization, and provide theoretical and experimental fundamention for the exploitment, improvement and improved variety selection of the germplasm of Chinese medicinal plants. Methods: The protoplasta of Panax quinquefolium L. was fused with that of Daucus carota var.sativus Hoffm. by PEG method. The regenerated clones callus, everyone derived form a single fused cell, were examined for their hybrids nature by morphology .Phenotype and Estenase isozyme , chromosome .RAPD and 5SrDNA spacer sequence analysis. HPLC was used in component of Ginseng Saponins Rbi and determined content of polysaccharose by Ultraviolet spectrophotometry method. Results: The rapidly growing carrot cell suspension systerms have been established; Have selected Radix Panacis Quinquefolii culture medium and subculture medium; There have been 29 clones in symmetric somatic hybrids between Radix Panacis Quinquefolii and carrot and ten clones were detected to be hybrids cell; The somatic hybrids had the characteristic bands of two parents and new bands in the electrophoresis pattern of Phenotype isozyme and RAPD; There have been preliminary heterosis that the content of Ginseng Saponins Rb1 of six in ten somatic hybrids are higher than that of in Radix Panacis Quinquefolii, two more prominence. Experiment suggested that the technique of somatic hybrids of Radix Panacis Quinquefolii and carrot have success.
