Panax quinquefolium L. is also named American ginsen, it belongs to Araliacas, Panax. P quinquefolium L. has been used as medicinal herbs for 300 years, the major functional component of P quinquefolium L. is ginsenoside. Panax quinquefolium L. has been cultivated for long time, but its growth period is very long, and it is sensitive to insect enviroment. and weather etc. the yield can not meet the need. One of ways that solve this problem is carrying out cell and tissue culture by using cell engineering. This paper studied the effects of some physical, chemical factors on the callus growth and saponin content in course of calli induced, solid callus culture and suspension callus culture with the young embryo of Panax quinquefolium L..The results showed:2,4-D and BA are better than other auxin and cytokinin for callus inducing, the most suitable concentration of them are 3mg/L and 0.5mg/L respectively; 23 + 1C,, dark. The callus inducing of Panax quinquefolium L.'s young embryo is the course of interaction of explant, growth factors, nutritional factors and environmental.MS + 2, 4-D2 mg/L + BA0.5 mg/L and 23C., dark are the best condition for solid callus culture. The growth curve turns on "s" appearance.Low concentration of 2, 4-D can improve saponin content; 2, 4-D associated with KT, saponin content can be improved obviously.Sucrose is more suitable for callus growth and saponin synthesis, the concentration of sucrose is 3%.MS+2, 4-D0.5mg/L+IBA2 mg/L+BA0.5 mg/L is the best condition for suspension culture. The growth curve turns on "s"apperance.The most suitable speed of shake flask is 106rpm, pH value is 6.07 and inoculum densities is 0.7g/20ml...
