Cucumber mosaic virus (CMV) is an important virus to tomato which can cause serious yield lost. The drug and the vaccine are not sufficient enough control the disease. Virus resistance breeding is the most econonomical and effective controlling method. Because of the lack of CMV resistance gene in tomao, traditional plant breeding ways cannot be efficient. Plant genetic engineering provides a new way for tomato's CMV resistance breeding. In this study we have developed some new tomato germplasms with CMV-dMP gene by Agrobacterium Tumefaciens.(1) CMV-dMP gene (K130) was transferred to Hufanl479 by Agrobacterium Tumefaciens. We develop new germplasms for tomato's CMV resistance breeding.(2) The factors that effect tomato gene transformation are analyzed. When the concentration of kanamycin is 100mg/L, the selection for transferred plant will be reliable. When the concentration of carbenicillin is less than 750mg/L, there will be no harmful effect to explants. Receptor genotype is an important factor that effect tomato gene transformation. Before transformation, we must choose suitable genotype. In this study we gain a tomato variety, which is suit to rapid reproduction and high efficient gene transformation. We are applying for a patent. Analyzing the different type of explants, we found that the tomato cotyledon explants have higher efficient than hypocotyls in transformation.(3) The results showed that the target gene has been inserted into tomato genome by kanamycin resistance expriment, and analyzing isoenzymes electrophoretogram, expressed the resistance to CMV by virus resistance test.
