Equine infectious anemia virus (EIAV), a member of lentivirus genus of retrovirus family, the similar to HIV on genetically and antigenically, causes an acute disease characterized by anemia, persistent infections, fever. Chinese equine infectious anemia virus donkey-leukocyte attenuated strain (EIAV-DLA) is the unique lentivirus vaccine which has been applied extensively for over 20 years in China mainland. The vaccine was developed by attenuation of donkey-adapted strain(EIAV-DA) in a unique donkey leukocyte cell culture system. The EIAV-DLA inoculated animals could resist challenges from homogeneous or heterogeneous virulent EIAV virus. In this study, we used different passages culture EIAV during the donkey-leukocyte attenuated. We extracted provirus DNA, amplified and cloned 5`LTR and 3`LTR of different passages culture EIAV strain. Sequence analysis shows that several variable genes related to virulent of virus. E-box motif, TAR, transcript-gene and poly(A) site have been found changes between virulent strains and avirulent strains. At the same time, it was the first time to find GATA motif in Chinese EIAV. This study gives an insight between LTR sequence evolution and virus attenuation.
