Study on transdermal absorbent effect and mechanism of enrofloxacin penetration in carpAzone was chosed as enrofloxacin permeating enhancer and transdermal absorbent mechanism of enrofloxacin transdermal was studied in the paper.This experiment included 9 groups. The marinating concentration of azone in group I,groupII ,groupIII,groupIV,group V,group VI was respectively 0.2 u 1/L-. 0.4 u 1/L> 1.0 u 1/L, 2.0 u 1/L, 3.0 u 1/L and 4.0 u 1/L, their immersing concentration of ERFX was 20mg/ml, The immersing concentration of azone in group VD was 0, and the immersing concentration of ERFX was 20mg/L. The immersing concentration of azone in group WI was 4.0 u 1/L, and the concentration of ERFX was 0. Group IX was blank control group. Drawing carp blood by the way of tail vein was proceeded in the point of 0.25h,0.5h,lh and 1.5h after immersing in the group I ,group II ,group,group III ,group IV ,group V ,group VI and group W, the enrofloxacin and its metabolite ciprofloxacin in plasma was analysized by HPLC. The skin and gill tissue was studied by histology and ultrastructural research to explore the permeation mechanism of azoneThe result was as follow: in the different azone concentration, the blood drug concentration of ERFX in the carp for groupIV,group V and groupVI was increased distinctly in any stage, and different significantly from groupVD(p<0.01).The ERFX blood drug in group I ,group II and groupIII after 0.25h was lower than group YD and distinguished significantly(p<0.01). After 0.25h, the finding of groupIII was higher than groupVE,and different evidently(pO.Ol), the concentration of group I and group II was still lower than groupVII ,but diversity wasn't obvious. After 0.5h, group II and group III were exceeded to group Vfl , and distinguished evidiently(p<0.01),the concentration of group I was still lower than groupVD,but distinguish wasn't evidient. After 1.5h, the ERFX blood drug concentration of group I was enhanced suddenly and exceeded to groupVfl obviously. The CPFX blood drug concentration of each group in the same time exceeded to group W,and distinguished evidiently.Observing by microscope, the very few branch leaf of group I ,group II ,groupIll,groupIV and group V produced curl lightly, respiration cells separated from bottom membrane to form a outside larger space. For 3 days after having terminated immersing , the histological strcucture of branch leaf was recoveried gradually, the change of curl became lightly, the outside space was smaller. 7days later, the change recoveried completely, the very few branch leaf of groupVI and groupVffl produced curl , respiration cells separated from bottom membrane to form a outside space which was irregular. 7days later, the histological strcucture of branch leaf was recoveried gradually, the change of curl became lightly, the outside space was smaller. After lOdays, the change recoveried completely. Comparing with control group, the interval of the epidermis cells of carp in trial groups became loose. The discrimination of microscopic intestines among all groups hardly existed.Ultrastructurally, capillary vessel of branch leaf in group I ,group II ,group III,groupIV and group V produced change in differerent degree. The clearance between pillar cell and breath epidermis cell broadened, breath epidermis cell membrane changed irregularly and curl lightly. The space between pillar cell and breath epidermis cell broadened significantly, the surface of membrane produced vacuolar in groupVI and groupVDI. In group I ,group II ,groupIII,groupIV and group V. The interval of epidermis cells and the fiber of corium layer became loose in vary degree in each group. The fiber in corium layer of group VI and group Vffl appeared rapture.Conclusion:1. The poper concentration of improving enrofloxacin absorption in plasma in carp was 3.0 P 1/L.2. When the range of concentration was from2.0 n 1/L to 4.0 u 1/L.in water, azone can incur lightly pathologic and fine structural change, and easily restored.
