| The inducing, subculture and the formation of flavonoid of callus in Acer Truncatum Bunge were systematically studied from the following aspects, basic medium, plant growth regulator, type of explant, installing method, sampling time, and culturing conditions etc. The results showed that: 1.The optimum conditions for the callus induction in leaves are as follows:MS medium supplemented with 2,4-D 2.0 mg/L,BA0.3mg/L, NAA0.5mg/L and PVP0.5% are suitable for the callus induction, the callus inductive rate can achieved 100% .The immature leaves collected from 2 year old or 10 year old tree from middle of March to April was the favorable for the callus induction.The optimal way of inoculation for the explants was that 0.5cm2 leaves'epidermis towards above and 1.0cm stem segment explants placed horizontally in the medium. The best pH value for the callus induction was 5.8.2. The optimum conditions for the leaf-drived callus subculture are as follows:Growth status of callus was better on B5 supplemented with BA0.5 mg/L, 2,4-D2.0 mg/L , NAA1.0mg/L and sucrose4%, the callus yield and increasing times were the highest ,it can reached 12.67 g/Flask and 12.3. The suitable pH value of medium are among 5.6~5.8.The callus cultured in light(1500Lux,12h/d) for 25d are suitable for the callus growth.3. The optimum conditions for the formation of flavonoid are as follows:The flavonoid content of the callus induced from the leaves that collected form10 year old tree in the period of the middle of May was the highest.In view of callus growth and formation of flavonoid, B5 basic midium was suitable. 4% sucrose was better for flavonoid formation of callus.The formation of flavonoid in callus need light, the content of flavonoid can reached 0.978% with the callus cultured in light (1500Lux,12h/d). The callus cultured in dark without of flavonoid.There were flavonoid in leaf-drived callus cultured in the pH value from 5.0~6.2, the flavonoid content was highest when pH was 5.6. The callus flavonoid yield was the highest value when the callus was cultured for 25d,it can reached 123.91mg/Flask. |
