Different Dioscorea Zingiberensis Populations stemming form Hubei and Shan xi provinces and individuals from Hubei-Yun xi Population were used as materials in the research. Genetic diversity and genetic relationships between different populations were analyzed by means of Polyaerglamide gel electrophoresis of peroxidase isozyme and esterase isozyme. Furthermore, the variety of the contents of proline, soluable sugar and MDA in leaves of different Populations and individuals were studied to understand their reaction and adaptivity to the heat stress. The main results are as follows:1. Dioscorea Zingiberensis Population POD and EST zymogram diversity degree prove to be high, 50% and 59%, respectively. Shanxi-Feng xian and Hubei-Song zi are geographically far and there hardly are gene exchanges between them. Therefore,. Differential coefficient of isozyme zyograms is the maxium and the genetic relationship between the two populations becomes the farest. However, Hubei-Yun xian and Hubei-Yun xi populations are geographically near and there are much gene exchanges between them. Therefore, Differential coefficient of isozyme zyograms is the minium and the genetic relationship between the two populations becomes the nearest. The research suggests that the Dioscorea Zingiberensis Population classification in terms of isozyme conform to the geographical chassification.2. The contents of proline and soluable sugar in leaves of different populations and individuals gradually change under heat stress. Different temperatures have different effect on the contents of proline and soluable sugar and they are higher under 45 C heat stress than 35C heat stress. In terms of the accumulation of Proline, Hubei-Jingmen, Hunan-Lu xi, Hubei-Shen nong jia, Shan xi-An kang and Hubei-Yun xi populations as well as H115, H100 and H94 show the greater adaptivities under heat stress. In terms of the accumulation of Soluable sugar, Hubei-Jing men, Hunan-Lu xi and Shanxi-An kang populations as well as H100 and H94 have the greater adaptivities under heat stress.3. The increase of the contents of MDA in leaves under heat stress showes that the cell membrane of Dioscorea Zingiberensis get injured. The higher the content of MDA is, the more badly it get injured. The different contents of MDA in different Populations and individuals suggest the different injures. The 45 C heat stress imposes the greater injury on the cell membrane.4. The contents of proline and the contents of soluable sugar in leaves of Dioscorea Zingiberensis populations and individuals have the identical variety tendency under heat stress and it means a certain correlation between them under heat stress.the correlation suggest tiie enzyme corrlelation. However, the contents of MDA have a different variety tendency, showing that the enzyme correlation does not exist between the variety of the content of MDA and the variety of the content of proline and soluable sugar in Dioscorea Zingiberensis under heat stress.
