Mature zygotic embryo of American ginseng (Panax quinquefolium) were used as explantto study the influence of different hormone and subculture on embryogenic callus occurrenceand growth, the explants were cultured on MS basal media supplemented2,4-D1mg/L and6-BA0.5mg/L, by subculture3times, loose and rapid growth embryogenic callus werescreened out. And then, by comparing different content combination of2,4-D and NAA, cellsuspension culture were established and free embryoid were obtained on MS+2,4-D0.5mg/L.Shaking4weeks,observed by a microscope, formation large of globular embryo andheart-shaped embryo. The embryoid bodies transferred to the medium MS+2,4-D0.5mg/L+GA35mg/L,After15days, formation large of torpedo-shaped embryoid period andcotyledonary embryoid,the optimum medium of Somatic embryogenesis is ight cultured4weeks, Free of embryoid formation. The best medium for the formation of Embryoid bodiesseedlings is1/2MS+NAA2mg/L+6-BA0.5mg/L+GA35mg/L, Of which, adding agarpowde6g. cultured2weeks, get a lot of American ginseng seedlings with structuralperfection and growth strong.Embryoid develops into seedlings. Using American ginseng (Panax quinquefolium)、Lilium brownii、saintpaulia ionantha materials, American ginseng Influences of differentventilating vessels on embryonic callus formation in mature zygotic embryo of three kinds ofexplants and its relationship with ethylene. Sealed with hermetically closed screw caps linedwith PTFE silica gel and ventilating plastic film.30days the result show:in medium ofventilating vessel embryogenic Mmature zygotic embryo of American ginseng﹑Lilium﹑Saintpaulia ionantha. The incidence rate of3kinds of explants were92.9%、94.6%、69.8%,respectively. the hermetically vessel were20.5%、64.3%、11.4%,ventilating vesselembryogenic callus rate has higher callus occur ability than the hermetically vessel.Ethyleneconcentration in the sealed vessel were4.3μL L-1、1.63μL L-1、2.8μL L-1, Due to the lowconcentration of ethylene can not be detected in the hermetically vessel.Observation on aregular basis and the ethylene in vessel were detected with gas chromatograph. Ventilating vessel embryogenic callus rate has higher callus occur ability than the hermetically vessel; thepossible causes of it was ethylene biosynthesis of self catalytic effect.
