OBJECTIVE: The nestling Muscovy ducks' high lethality has been found during these years when they were infected by Muscovy duck reovirus(DRV).According to the clinical symptoms and related document data, we conclude the receptor of DRV is probably the receptor of P 2-adrenerlin(p"2-AR). The molecuLar weight of DRV's receptor wouLd be identified in this paper and compared with that of MRV-III's receptor which is just the receptor of ?-AR so as to find whether they are identical. On the other hand, the variation of the ?-adrenalin relasing concentration following the symptoms' developing wouLd be also checked after the Muscovy's being infected by the DRV so as to identify whether they are intensely related between the DRV's receptor and the ?-AR. All that wouLd be done would offer us further proof to confirm what the DRV's receptor is. METHODS AND RESULTS: Propagating the virus by inocuLating DRV B3 strain to the vigorous Muscovy duck fibroblast, Collecting the virus when the cytopathic effect(CPE) is up to 50%~80%, freezing and thawing the virus solvation for three times , condensing it by centrifugal and purifying the virus by ammonium suLfate precipitation and dialysis , checking the concentration of the virus protein by uLtraspectrophotometry, the resuLt is 5mg/mL. The titer of the serum from the four inocuLated stout Muscovy duck was about 1: 32 checked by the agar gel diffusion experiments.The concentration of the purified erythrocyte membrane protein was checked which is 29mg/mL. The protein bands were the most distinct under the conditions such as the sample with the concentration of the membrane protein 7.5ug/uL and of the marker 0.4ug/uL, the sample volume used for one hole at a time 50uL and constant electric current 30mA. The molecuLar weight of the virus' receptor 72kDa was checked out by statistics software SPSS with the help of the molecuLar weights and the mobility of the marker protein after the receptor is showed by VOPBA which is to say that the membrane protein reacted with virus ,serum and enzyme-linked antibody II in turn after all of them were transferred from the gel to PVDF membrane. The releasing concentration of ?-adrenal ine keep increasing in the first three days and decrease later after the ducks were inocuLated DRV.CONCLUSION: The receptor of DRV is similar to the ?AR. Maybe both are the same membrane protein.
