In Vitro Maturation And In Vitro Fertilization Of Oocytes From Slaughtered Goat

In order to abtain plenty of high quality and low-cost early embryos, the study of IVM and IVF of slaughtered goat's oocytes were done.Goat ovaries used in the research were collected from slaughter house. Oocytes were collected with aspiration and slice.The study compared the influences of addition of 15%FBS, 10%FBS, 20%ENGS and 15%ENGS on IVM of goat's oocytes.Maturation rate was 61.12%, 64.49%, 72.37% and 67.62% separately.The maturation rate of culture media 1 was high significantly lower than those of culture media 3 and 4.Maturation rate of culture media 3 was high significantly higher than that of culture media 2,significant higher than that of culture media 4.Goat'oocytes were cultured at 37C, 38.5C, 39C to study the influence of temperature on IVM,.Maturation rate was 50.00%, 65.66%, 71.15% respectively.The result showed that the maturation rate at 37 C and 39 C were high significantly different,the maturation rate cultured at 37C and 38.5Cwere different significantly.The effects of culture time on goat oocytes IVM was studied.Oocytes had been cultured for 18h, 20h, 24h, 26h.The maturation rate was 53.22%, 59.94%, 71.70% and 72.12% respectively, the fertilization rate was 48.22%, 56.70%, 60.56% and 61.12% respectively.The result showed that the maturation rate when oocytes cultured for 24-26h was significantly higher than others, and the fertilization rate was high significantly higherthan that of cultured for 18h.The fertilization rate of IVM oocytes was 62.5%, and fertilization rate of oocytes after superovulation was 65.0%, the latter was little higher.The large antral fillicar oocytes and small antral folliclar oocytes were collected and cultured respectively. The result showed that the maturation rate and fertilization rate of large antral folliclear oocytes were 71.74% and 60.16% respectively,high significantly higher than those of small antral folliclar.
