The stable, efficient plant regeneration system is prerepuisite to success for transformation with Agrobacterium tumefaciens. In this study, the shoots isolated from kiwifruit (A. chinensis var. rufopulpa Liang et Ferguson) were used as explants. By means of the orthogonal design, analysis of variance and multiple comparisons, the factors, including basic media, plant growth regulators, seasons, were analyzed and the ideal regeneration system of kiwifruit (A. chinensis var. rufopulpa Liang et Ferguson)was established. The results were as follows:(1) The method to get clustered buds from apical and lateral buds was not so good, and it was given up while the method to get regenerated buds from callus was kept down.(2) To get regenerated buds from callus, the stem without epidermis was the optimum explant and the most fitting season to gather the stem was summer. To compare the differentiation of different callus from stems, petioles and leaves, the differentiation rate of callus from stems was 83.73%. However, it was 12.88% from petioles and leaves only 8.10%.(3) Take stem as explants, the MS medium was more suit as basic medium than the others. On the stage of callus induction, the MS medium supplemented with 4.0mg/LBA and 4.0mg/L IAA was the best one, with regeneration rate of 61.24%. On the stage of buds differentiation, the best medium was the MS medium containing 2.0mg/L BA plus 0.3mg/L IBA with bud regeneration rate of 88.82%. In the root regeneration trial, 1/2MS medium with 0.2mg/LNAA was more benefitial than the others. To get more cuvette plants, the bestmultiplication medium was the MS medium containing 2.0mg/L BA plus 0.3mg/L IBA with the multiplication rate of 5.32.(4) Take the leaves of cuvette plants to regenerated buds, the optimal initiation medium was the MS medium supplemented 2.0mg/L BA and 0.2mg/L NAA. And the bud regeneration rate was 87.03%.(5) The regenerated plants with roots were planted in the medium containing 25% vermiculite, 25% perlite, 20% sand and 30% soil with the survivor rate of 90% after a month.
