In this study, the genetic analysis on HMW-GS was evaluated. The variation about the gliadin and the SSR were detected by the way of the A-PAGE and clustered for analysis.1. The composition of HMW-GS was evaluated by sodium dodecyl sulphate polyacrymide gel electrophoresis (SDS-PAGE) in 123 species collected from the west and north of china. A total of fourteen alleles and sixteen subunits combinations were identified, and 5 of these subunits were found to be different from those previously detected by Payne and Laqrence in hexaploid wheat. The results indicated: (1) The frequency distribution of 2* subunit in tetraploid wheat was higher subunits in Glu-A1 a than hexaploid wheat. (2) The variation of HMW-GS in tetraploid wheat was very rich. The compositions of Glu-B1 sites were different obviously from that of hexaploid wheat, dicoccum and durum. This study suggested: tetraploid wheat, the wild relative of wheat presenting high polymorphism of the HMW-GS and subunit composition, was an utilizable resource for broadening the genetic basis for wheat breeding.2. Gliadin variations were detected among 98 tetraploid wheat. The gliadin patterns were similar to each other among of the relative species or the closer distribution of geography. In a, b, r and to zones, the gliadin band combination were different among each near accessions. For example, between AS2295 and AS2296 from Sichuan only one the gliadin band is respectively different in a, B and w zones. That of AS2296 and AS2300 is only one band in the y and w zones, respectively.3. In SSR marker analysis, 16 primer located on 9 chromosome arms, couldreveal rich polymorphisms among 48 species. A total of 97 SSR loci were detected. 2 to 14 alleles were detected at each locus, with an average of 6.1. The most of locus detected is 14, from the primer GWM162. While the primer GWM24 could only detected 2 loci. The polymorphism distribution showed the rich diversity. The cluster analysis the variation of SSR locus could be caused by environment difference.
