Thinopyrum intermedium (Host)B.D.,a hexaploid species(2n-6x=42,StElE2),is an important source for improving the genetic variability of cultivated wheat .Desirable characteristics of Th. intermedium of potential valuable for wheat improvement include perennial habit ,stress tolerance and disease resistanceIn order to transfer the genes of disease resistance from Th. intermedium into Triticum aestivum(2n=42,ABD) particularly, the hybrid progenies between T. aestivum and Th. Intermedium Partial amphiplodis ZhongS were screened. T. aestivum- Th. Intermedium Disomic addition and substitution were identification with the assistance of Meiotic M I chromosome preparations, mitotic chromosome C-banding, Genomic In Situ Hybridization(GISH),.Two study material is 011077, from zhong 5/"s"265 and Ols-20, from zhongS /"s"265 //chuan yu 12 .the main results as follows:1 .011077, Ols-20 are resistance to stripe rust .cytogenetic analysis showed that chromosome number in roop-tip cells of 011077 is 44 ,including six satellite chromosomes, with an average chromosome configuration of 0.481+21.4811+0.19IIIin Percell characterized meiosis of its pollen mother cells. Ols-20 is 42 chromosomes ,four satellite chromosomes.The observation of chromosome pairing at metaphase I in PMCs of hybrids of Ols-20 with chuan yu!2,FI average chromosome configuration is 21+19.9911.These suggest that Ols-20 is a disomic aline substitution or translocation of big segment from Th. Intermedium2 Giemsa-C banding showed the two have two chromosomes with Th. Intermedium character banding in telomeral..3 In GISH using the St genome as a probe ,common wheat genome as a blocker indicate that a pair of Th. Intermedium St chromosomes substituted for a pair of wheat chromosomes in Ols-20;while in disomic alien addition line 011077 has a pair of St-W chromosomes and the St chromosomes have satellites belonging to homoeologous group 1.
