In this study, hyaluronic acid (HA) was added to the modified foundation medium , throughwhich to mainly determine the effect of HA on in vitro maturation (IVM) of bovine oocytes,in vitro fertilittation ( IVF), in vitro culture (IVC) Freezing Conservation (FC).The resultshow that: 1. the group with 4mg/mL HAwas significant higher than other groups in the frequencyoffirst polar body extrusion (PB) and cleavage rate of oocytes(P<0.05), this primitivelyexpresses that 4 mg/mL was the best concentration for free serum culture with HA. 2. When we added HA , bovine serum albumin (BSA)or Oestrus Calf Serum(OCS )toTCM-199, the group with the OCS was significantly higher than other two groups (83.40%,84.33% and 92.17%, P<0.05) in PB, but cleavage rate was no significant different in threegroups P>0.05; there were no different in PB of BSA and HA (P>0.05).Thereof HA was asubstitute for bovine serum albumin(BSA)in serum-free medium to mature the oocytes,butthe effect was not equal to blood serum in result of the culture. 3.HA in TCM-199 and Synthetic Oviduct Fluid (SOF) have no effect on in vitromaturation of bovine oocytes in PB of oocytes and cleavage rate; it demonstrated that HA canfavor the maturity ofoocytes in complicated or simple free-serum medium. 4. It is important the effect of the concentration of HA on bovine earlier embryodevelopment, to or not add HA is significant different (P<0.05) between groups; whenconcentration of HA is 2~4 g/L, there is 31.76% of blastocyst rate, which show 2~4 g/L is thebest concentration. 5. In different additives, the group with HA demonstrated significantly higher than groupwith polyvinyl- alcohol (PVA)(respectively for 31.54% with 26.61%,P<0.05)in blastocystrate,but lower group with FCS (P<0.05); it was no significant different with BSA group (P>0.05) in blastocyst rate; Those which show BSA will be best replaced by HA. 6. In free-serum medium, oocytes matured with HA is significantly higher than PVA inblastocyst rate (respectively for 32.35% with 26.80%, P<0.05); However, it is significantlylower than ECS (respectively for 32.35% with 34.91%, P<0.05); Although it is higher thanBSA there is no significantly different in them (P>0.05). 7. Although Synthetic Oviduct Fluid (SOF) is higher than TCM-199 there is nosignificantly different in them (respectively for 31.03% with 30.43%, P>0.05), which showHA can all improve bovine embryo development in SOF and TCM-199. 8. Embryo cultured by TCM-199+ HA was significant higher than group that did not addHA (P<0.05) in its survive rate and hatch rate (respectively for 65.85% vs 58.69% and60.98% vs 36.96%) after freezing-thawed. 9. Embryo cultured by SOF+ HA was significant higher than group that did not add HA (P<0.05) in its survive rate and hatch rate (respectively for 58.33% vs 43.14% with 52.08 vs31.37%) after freezing-thawed. 10.Embryo cultured by TCM-199+ HA was significant higher than SOF+HA in survive rateand hatch rate (P<0.05) after freezing-thawed.
