Bovine mastitis,especially subclinical mastitis is high frequency disease in dairy whichcan cause heave economical loss.Alouth researcher work hard all over the world.there is nogood method to control it.The most control method is using antibiotics.It is necessary tocreate new method in order to control mastitis,because antibiotics not only have byeffect,also it can decrease milk quality.Certainly,vaccine prevention is the best way forcontrolling.But till today,only E.coliform J5 vaccine is successfully development,itspretection ratio is more than 85%.But the dominant pathogens are G+ germs in our country,so it is important to development G+ germs vaccine.Our goal is to investigate and isolatedominant pathogens of mastitis in the area nearby xi'an and development G+ germs vaccinebasis on those work all over the worldSCC is thought as a standard method in monitoring milk quality in many countries,because itis closely related to IMI. The regular method is hard to practice owing to fat inmilk.especially to colostrums and ending milk.This report introduce a new method which isinduced from the regular method.This new method is simple and convenient for practise.Byusing this new method,we found some facts that were not reported in the past.1 Make progress in SCC methodSCC is thought as a standard method in monitoring milk quality in many countries,because it is closely related to IMI.The regular method is hard to practice owing to fat inmilk.especially to colostrums and ending milk.This report introduce a new method which isinduced from the regular method.This new method is simple and convenient for practise.Byusing this new method,we found some facts that were not reported in the past.2 Isolation and Identification of Bovine Subclinic mastitis pathogensWe have investigated infection ratio of dairy subclinical mastitis in xi'an area of Chinasince Oct.2002.We got 48 samples from 1000 cows and diagnosed those total 184 quartersby LMT and SCC. The results revealed cow infected subclinical mastitis total to62.7%.Quarters infected sublinical mastitis total to 34.2%.Cow uninfected total to61.2%.subclinical mastitis only total to 16.7%.We isolated 67 strains of pathogens including41 strains of Staphylococcus,total to 61.2%.20 strains of streptococcus ,total to 29.9%.3strains of G+ bacteria,total to 4.5%.3 strains of G- bacteria ,total to 4.5%.3 strains of G+ bacteria were identified as bacillus thuringiensis.After a series of tests,we think bacillus thuringiensis as one cause of bovine mastitis. We also found its cultureafter 24h in broth can cause mouse death by intraperitoneal injection.this is different fromothers'reports.3 vaccine development and evaluation3.1 Immunization with lysate bacteria isolated from mastitis bovine milk on rabbit4 kinds of predominant G+ bacteria isolated from mastitis bovine milk were used toprepare lysate vaccine mixed with 3 sorts of adjuvants. After immunization, the rabbitsblood samples were collected every 7d from 7th day postimmunization. In order to determineif these 4 kinds of predominant G+ bacteria have better antigenicity and select properadjuvant, we collected sera to determine agglutination titer by agglutination reaction test tube.The result revealed, agglutination titer have arised after immunization and increased to peakabout 28th day postimmunization. It sustained at high lever at 63rd day postimmunization.Adjuvant C are the best among these 3 sorts of adjuvants, but there aren,t difference instatistics. Compare to control group, all these 3 kinds of adjuvants are significant differencein statistics (P<0.01)3.2 Immunization with lysate bacteria isolated from mastitis bovine milk on milk goat4 kinds of predominant G+ bacteria isolated from mastitis bovine milk were used toprepare lysate vaccine mixed with 3 sorts of adjuvants.
