In the study, Spreqne-Dawley rat was chosen as in vivo model to examine interleukin-1beta (IL-1β) expression level in uterus and placenta in the different phase of pregnant and the effects of interferon-gamma(IFN-γ) and IL-1β on the expression of IL-1β in uterus and placenta using RT-PCR and immune histochemitry. The results were as follows:(1)In the course of pregnancy, uterus and embryo expressed IL-1β, which changed according to time and order. Maternal decidual cell had a little IL-1β in early gestation. Decidual rebuilding vein and maternal-embryo interface expressed more IL-1β and part syncytiotrophoblast also expressed more IL-1β in mid gestation. Decidual rebuilding vein and trophalast cell including syncytiotrophoblast and cytotrophoblast expressed some IL-1β. (2)IL-1β expression in uterus was significantly reduced in peri-implantation,post- implantation,mid and late gestation period, and IL-1β expression in placenta was significantly decreased in late gestation period after injecting over-dose IFN-γ. This indicated over-dose IFN-β could suppress the expression of IL-1β of uterus and placenta in certain pregnant phase.(3)IL-1β expression in uterus was significantly increased in peri-implantation,post- implantation and mid gestation period, and IL-1β expression in placenta was significantly increased in early and late gestation period when injected over-dose IL-1β. This indicated IL-1β could prompt the expression of IL-1β of uterus and placenta in certain pregnant phase.(4)IFN-γ restrained development of embryo and placenta through decreasing the expression of IL-1β, and induced abortion and premature. The effect of IL-1β on development of embryo and placenta was different in the different phase of pregnancy.In summary, the expression of IL-1β in uterus showed evident trends, and in placenta had little change. In certain pregnant phase, IFN-γ could suppress the expression of IL-1β of uterus and placenta, while IL-1β could prompt the expression of IL-1β. This shows Th1 type cytokines are always not to induce the expression of homotype cytokines.
