Samples of diseased scallop (Chlamys farreri) were collected from taping Bay ofQingdao, in the period of death in large scale from 2002 to 2003. Sucrose gradient centrifugation , transmission electron microscopy technique of negative staining and SDS-PAGE were applied to study the purification , internal structure and protein compositions.1. The virus-like particles of scallop (Chlamys farreri) were purified successfully by means of sucrose gradient centrifugation. According to the TEM test, virus bands existed in three surfaces among different sucrose layers: 32.5-35%surface , 35-37.5% surface and 37.5-40% surface. On the base of comparing observation on the morphology and structure ,depending on the features of taxology, it can be concluded : 1) the virions in 32.5-35% surface , accounting for 82.5% in number, are Paramyxoviridae virus particles;2)the virions in 35-37.5% surface , accounting for 88.5% in number, are Coronaviridaevirus particles; 3)the virions in 35-37.5% surface , accounting for 1.3% in number, areBunyaviridae virus particles; 4) the virions in 375-40% surface , accounting for 0.9% in number, are wwsv particles;2 . Super-micro structures and protein components of Coronavirus-like particles are observed and analyzed through the techniques of negative staining and SDS-PAGE. A structure model of the virions is built on the base of comparison with other Coronavirus .Two ICS proteins are identified , referring to molecular mass and site of other Coronavirus structure proteins .The 42.5KD protein is the M protein ,which is a primary composition of ICS membrane. The 58KD protein is N protein. Two spike proteins are identified . The 151KD protein is S protein, and the 60KD protein is HE protein.3 . Super-micro structures and protein components of Paramyxovirus -like particles are observed and analyzed through the techniques of negative staining and SDS-PAGE. On the base of the comparison with the structures and compositions of other Paramyxovirus ,themodel structure of scallop (Chlamys farreri) virus is discussed . The relation between the spherical particle and filiform is also discussed. Two proteins of spike and one protein of nucleocapsid are identified. The 62KD protein is NP The 74KD protein is HN protein. The 56KD is F protein.
