The orchid family is a very important plant resource. It has huge economical value and become an important part in the flower industry. And also most of them have very high medicinal value with huge market. Orchid mycorrhizal fungi, which offer nutrition for orchids and promote its growth, can form symbionts with orchidaceous seed, corm and nutritional root. In this paper, wild Changnienia amoena Chien mycorrhizal microstructure and fungal intrusion way were studied. Furthermore, the fungal strains isolated from wild Changnienia amoena roots were identified and reinoculated. In additon, fungi fertilizer was made and applied in promotion orchid growth. The results are the following:1. The mycorrhizal microstructure of wild Changnienia was the typical mycorrhizal structure of orchids. Mycorrhizal fungi enter the velamen and exodermis through root hair and invade the cortical tissue by the exodermal passage cells, and then form pelotons.2. 36 fungal strains have been isolated. Isolates were identified on the basis of colonial morphology and microscopic characteristics of hyphae, spores, conceptacle, number of karyon and fruitbody. Generally, these were shown to be members of the Basidiomycota and Mitogenic fungi. The superior genus is Fusarium, the other genera each belong to Ceratobasidium, Epulorhiza, Epulorhiza repens, Monilipsis Ruhland(1908), Sitostrema, Cephalosporium, Cylindrocarpon, Geotrichum, Oidiodendronand Paecilomyces.3. After five isolates inoculated to seeds of Cypripedium tibeticum, C. bardolphianum, C. flavum and Calanthe delavayi, there are a few seeds of C. bardolphianum germinate with strain (DHL-13).4. Sixteen isolates were inoculated to tissue culture seedlings of Dendrobium sinense. In medium I , ten of them were effective on promotion growth at different level after inoculation. After inoculated with isolate DHL-19, the average height increment of the seedlings is more significantly higher ( α =0.05) than the comparison seedlings. And also inoculated with isolates (DHL-16, DHL-17), it was most significantly highest ( α =0.01) than the comparison seedlings. The inoculated isolateswere reisolated from the seedling roots and also small pelotons were found. So, DHL-16, DHL-17 and DHL-19 should be the mycorrhizal fungi of orchid.5. Fungi fertilizer was made and applied to the roots of Paphiopedilum concolor. The result showed that the average flower number of treatment groups remarkably exceeded the comparison group, and the treatment group I was more significantly higher than that of the comparison group ( α =0.05). So, the fungi fertilizer was effective on promotion the growth of orchid. It also promoted flowering of Paphiopedilum concolor.This study was a good try. It showed that fungi isolated from wild Changnienia amoena roots had great value in the protection, multiplication and usage of wildChangnienia amoena.
