| Objective: A static tilmicosin liposome was prepared successfully and firstly by revolved-evaporation method. The study of its distribution in plasma and tissues of chicken provided the theoretical basis for veterinary investigation and clinical using. Methods:(l) Making the liposomes with the improved method, and examining its shape size coefficient and stability; (2) 336 chickens were divided into three groups and injected in breast muscle with TMSL, TMS, water. Sampling blood,muscle,kidney,lung and liver at 0.5, 1.0, 2.0, 4.0, 8.0, 12.0, 24.0, 46.0 and 72.0h after the injection, then determining the concentration by HPLC. Results: (l)Its shape was round yellow perspective liquor and its size was determined by dynastic light scattering. The mean size of liposome was 98.6 run; almost 80% of them were between 50nm to 150nm. The closed rate which detected by Spandex G-50 gel method ranged from 90 to 92 %( n=3), the production stability preserved more than five months at 4℃; (2)The result of plasma determined by HPLC showed its long action. The chicken plasma of two group were given t test,0.01< p<0.05, the purpose was notability, and the polarity drug concentration of TMS group in plasma was less than the TMSL group; (3) The results of tissues determined by HPLC showed its target. The drug distribution of two group in muscle, kidney, liver and lung were given t test, the results as follow:I . TMS group: The drug concentration of liver was most , then following were lung, kidney and muscle , their retention time was 16h, 4h, 2h and lh respectively.II. TMSL group: The order drug concentration was still like upside, but the amount of them were more than before ,and the retention time were 20h, 8h, 8h and lh, respectively.The comparative result of two group showed that the lung and kidney were notability target organ, 0.01 |
