Abstract/Summary:
In this study, The E2 gene nucleotide sequences of Prevalent Virulent Strain of classical Swine Fever (CSFV) collected from Inner Mongolia were amplified by reverse transcription polymerase chain reaction (RT-PCR) and nested polymerase chain reaction (nPCR), then, the amplified fragments were cloned in PMD-18-T vector and sequenced. The nucleotide sequences and the deduced amino acid sequences of the Inner Mongolia strain (NMFL) and C-strain were compared and analyzed by DNAstar software. The further study of genetic relationship between the major protective antigen encoding regions of E2 genes of Prevalent Virulent Strain and CSFV C-strain was done by constructing the phylogenetic tree . The results showed that : 1 The E2 gene of NMFL strain and C-strain were all 1170bp in length , they contain total signal peptide sequence and part transmembrane domain structure sequence. 2 The homology of NMFL strain and C-strain were lower which were 82.6% for nucleotide sequence and 87.9% for deduced amino acid sequence. 3 The NMFL strain and C-strain were classified into different subgroups in the phylogenetic tree because of their lower homology , meanwhile, the homology of NMFL strain and other reference strains were high ,so they were classified into group 2 in the phylogenetic tree, and the C and SHIMEN strain were classified into group 1 in the phylogenetic tree.
