Stylosanthes spp.is the most important commercial forage legume in tropical areas of the world. However, it was affected severely by the stylo anthracnose. It is a new way to breed new disease-resistant varieties of the Stylosanthes spp.by transformation and expression of the disease-resistant genes into Stylosanthes spp.A rapid and efficient regeneration system of Stylosanthes guianensis cv. Reyan No.2 was established and optimized by comparing the effects of different hormone combinations on the induction of callus, regeneration of adventitious shoots and roots. Than the fusion gene constructed with chintinase gene and P -1,3-glucanase gene was introduced into Stylosanthes guianensis cv. Reyan No.2 by using the Agrobacterium-mcdiated transformation method. Thirty kanamycin-resistant transformants were obtained and eight kanamycin-resistant transformants were checked subsequently by PCR and Southern blot. There are three transgenic plants the analysis of the PCR and Southern blot suggested that the fusion gene was integrated into the genomes of three transgenic plants.
