Liquid dog semen preservation and cryopreservation was researched, and extenders were selected, then, the quality of frozen-thawed dog semen was assessed. In test one, the best preservation temperature and extenders in liquid dog semen were searched , and the available methods in dog semen cryopreservation were studyed. In test two, the quality of frozen-thawed dog semen was evaluated in different aspects.The results of the test one showed that liquid dog semen could survived longer at the temperature 15℃ than any other groups(P<0.05), and the extender No. 3 was the best for diluting dog semen in liquid semen preservation tests, there was significantly different compared with control group (extender No. 1) (P<0.01), while extender No. 2, 4 and 5 were better than extender No. 1(P<0.05). In the test of dog semen cryopreservation, extender No. 5, 3and 6 were greatly better than extender No. 1(P<0.01), and extender No. 4, 7and 8 were better than extender No. 1(P<0.05). In the test two, the method of frying in liquid nitrogen had better result than suffocating over liquid nitrogen. After thawed, the pellets were better than straws (P<0.05) and ampules (P<0.01). The quality of frozen-thawed semen was assayed with the motility recovery rate (MRR), the zona-free hamster ova penetration (SPA), the hypoosmotic swelling test (HOS) and the electron microscopic observation, etc. The result showed there was significant consistent between HOS & SPA. Significant correlation were observed between HOS & MRR (r=0.94, P<0.006) and SPA & MRR (r=0.93, P<0.049). The quality of frozen-thawed semen in different extenders were compared, and the effect factors to dog semen cryopreservation were analysised. The result showed that sugar was very important in dog semen cryopreservation, and the role of raffinose was better than sucrose,fructose and glucose. The electron microscopic observation revealed that sperm head plasmalemma and acrosome were the most cryosensitive.
