Semen preservation technology is very important for boar artificial insemination, while the quality of boar semen diluents determines the effect of preservation. The technology of boar semen normal temperature preservation in abroad is ahead of us, researches on the diluent formulation ingredients and the ratios are more in-depth than ours. However, our researches and developments in the dilution powder started late. Now, Most of Chinese pig raising enterprises and farmers depend on imported dilution powder for boar semen normal temperature preservation. It is not conducive to the healthy development of pig industry in China.Therefore, research and development on good quality diluent formulation will have great significance for reducing the cost of pig raising, increasing the pig breeding efficiency, and promoting the development of pig breeding industry in China.In this test, the mechanism of boar semen normal temperature preservation was studied, while there were a lot of researches about the protection mechanism of different substances for boar semen. Effects of GSH and SOD on boar semen preservation were studied,then we developed three kinds of formulas for boar semen diluted powder, and tested the dilution powders by the study of boar semen normal temperature preservation. Finally we screened out the best diluted powder of the three and test it’s effect in practical production compared with commodity diluted powder. The results were as follows:1. BTS with 1 mmol/L of GSH could reduce oxidative damage on sperm, decreased the content of MDA、H2O2 in semen(P < 0.05), and could significantly improve the SOD activity of sperm(P < 0.05); There is no significant difference(P > 0.05) in the effective preservation time and motility of sperm when 1 mmol/L and 5 mmol/L GSH was added in Modena. Both of the sperm effective preservation time were 5 days, and the sperm motility was 0.60 and 0.63.The results showed that the optimum adding amount of GSH is 1 mmol/L.2. The results showed that the addition of 200 IU/mL SOD in Modena could significantly improve sperm motility of boar, and reduce the content of MDA and H2O2 in boar semen and enhance the SOD activity in semen(P < 0.05), The sperm motility after stored 4 days was 0.657. While the addition of 300 IU/mL SOD in BTS could significantly improve the effect of boar semen ambient preservation, the sperm motility after stored 4 days was 0.653. The right amount addition of SOD could improve the preservation of sperms, would it will produce toxic effects on sperms if the addition of SOD excess.3.The effect of preservation was the best when sperms were diluted with formula III, the sperm motility after sperms stored 5 days was 0.60. There is no significant difference(P > 0.05) in the effect of preservation when sperms was diluted with formula III and Modena. There is also no significant difference(P > 0.05) in the effect of preservation when sperms were diluted with formula II and BTS. Both of their effective preservation time are 4 days. The effective preservation time of formula I treatment group was the shortest. But even so, the sperm motility of the formula I treatment group was excellent, and it would last 1~2 days. There is no significant difference(P > 0.05) in all indicators when sperms were diluted with those 5 kinds of formulas in 1 day. After 1 day of storage, plasma membrane integrity rate and acrosome integrity rate in Modena, formula I and formula III treatment group were significantly higher than in BTS and formula II treatment group(P < 0.05),and the plasma membrane integrity rate and acrosome integrity rate in formula I treatment group was the highest. But after 2 days of storage, the plasma membrane integrity rate and acrosome integrity rate in formula I treatment group decreased fast, were significantly lower than those in other 4 kinds of formula treatment groups. After 3~5 days’ storage, plasma membrane integrity rate and acrosome integrity rate of boar semen stored in Modena and formula III were significantly higher than that in BTS and formula II(P < 0.05)4.The diluted powder formula III was used to test the conception of Yorkshire、Duroc and Landrace in this study, The application effect of the formula III is excellent for Large White and Landrace, while the conception rate was more than 90%.There is no difference in application effect between the formula III and some famous commodity formulas. It proved that the formula III can be used in the practice of production.
