This paper is about flax (L. usitatissimum) genetic transformation by Agrobacterium tumefaciens mediated NP-1 gene. The transgenic plants express NP-1 gene steadily and be proved resistance to Fusarium Wilt.Research approach is to use 7309 variety as material, to base on tissue culture and Agrobacterium tumefaciens mediated, to build genetics transformation system, and we successfully gain transgenic ilax plants. We inoculate pathogen with flax plants through draw line and spray methods, to measure the ability of resistance disease. We draw a conclusion that the genetic flax improve the ability of resistance to Fusarium Wilt.In this research, we compare several methods that effect on tissue culture and genetic transformation in flax. First, we use hypocotyls segment as explant to preculture in different stages, it will impact on rate of resisting flax plants. The result shows: we use hypocotyls segments from sterile seedling that sprout in three days, and culture them about three days on B5 Medium with IAA 4mg/l and 6-BA 2mg/l to induce their callus and wound their callus. It will improve the rate of regenerated shoots. On the contrary, the rate of regenerated shoot is very low. Secondly, we compare antibiotics both Kanamycin and G418 as selective pressure. The result shows: callus is more sensitive to G418 than Kanamycin. When we use B5 medium with G418 15mg/L, we can get resistance shoots effectively.Through regeneration root of shoot, we gain sixty plus regeneration plants, then, transplant them to soil in greenhouse, survive fifteen regeneration plants, the number of their seeds are from fifteen to fifty. We identify next generation through PCR and PCR-Southern blotting, there are six positive plants. Further, we identify these plants in transcription level through Reverse Transcription-PCR, two plants have different expression at mRNA level. Through inoculate Fusarium oxysporum on the transplant, serial number NP6 and NP12 can enhance resistance against Fusarium Wilt.
