| In this article, an effective and accurate double selective system consisting of Nicotiana glutinosa necrotic type numeration and Nicotiana tabacum cv. NC89 TMV ELISA is constructed for pesticide selection against TMV. By selection of over ten kinds of chemicals and plants extracts, BABA and DEM are highlighted as 2 promising anti-TMV chemicals. Further research suggests that the most effective treatment of BABA is foliar spaye 3 days before TMV infection at a concentration of lmM; the optimal work concentration of DEM is 100MM. They can reach the suppression rate at 38.1% and 53.8% respectively.The passivation test of TMV in vitro shows that BABA's anti-TMV effect may largely depends on its induction of plant defence system rather than passives TMV directly. 2 different expressing gene segments, which are modified by BABA application, are discovered via DDRT-PCR technology. Segment 4A51, one of the two segments, shows reduced expression between 12~36h post BABA application. Nucleotide blast illustrates that it has very high homogenity with S in Petunia inflata.. S is a part of S locus and has the function of detecting the specialty of male gene and control self-incompatibility. The specific function of this segment, however, is unclear. The expression of the other segment C31 is promoted between 12~48h post BABA application, while no homogenous gene segment is found in nucleotide blast. |
