Abstract/Summary:
The study and extension of Miscanthus giganteus, a kind of gramineous high grass with great potentiality as ornamental and energy plant, is heavily limited by its natural sterility. To solve the above problem, propagation methods and adaptability of the micropropagated seedlings of Miscanthus giganteus were studied in this study, and main results as follows: (1) The micropropagation system through axillary shoot was established, including detaching the nodal segments by about 15 mm at 5-6 leaves stage, adding 6 mg/L 6-BA and 0.25mg/LNAA into induction and subculture medium, adding 5g/L active carbon to root induction medium. 100 and more seedlings could be produced from a single seedling in half year, followed the above protocol. (2) Another micropropagation method through embryoid regeneration using unemerged florescence as explant was established. The optimum callus induction medium was MS medium containing 2 mg/L 2, 4-D and the best regeneration medium was MS medium adding 5 mg/L6-BA and 0.25 mg/L NAA. Applying the above system, about 300 seedlings could be produced from a middle length florescence in half year. (3) Cuttage system was reported firstly. Sampled the shoot cuttings in July, divided into segments with one node, planted segments in field after water soaking for 4 days under 25℃,then 160 rhizome-shoot could be obtained from one. (4) The main characteristic of the above three methods and conventional propagation through rhizome-shoot were compared. The efficiency of the propagation through rhizome-shoot was highest, the input of cuttage propagation is lowest, while micropropagation through axillary shoot need small number maternal rhizome and was less limited by time or space. (5) For the Miscanthus giganteus seedlings to grow well, the most suitable temperature was 30℃, the NaCl tolerance limit was 0.8%. and the lowest water field capacity was 45%.
