This paper studied on the degradability of wheat gluten protein in goat's rumen, gluten protein on the movement of rumen of goats , the effect of gluten protein on ruminal metabolism and blood indexes in goats and the rumen fluid with opioid activity on the growth and endogenous hormones of local chickens. Also, activities of opioid peptides from gluten protein by the degradation of rumen microbes was determined with determination device of intestine of rabbits by culture in vitro. All these studies provided theoretical and experimental backup for exploring wheat gluten protein. The research includes three parts as follows:Part â… I : Studied on degradability of gluten protein in the rumen of goatsThe ruminal degradability of wheat gluten protein was determined by nylon bagstechnique with four rumen-fistulated goats. The Degradability of wheat gluten protein at 2, 6. 12, 24, 36, 48, 72h were 18.95%, 24.49%, 35.18%, 49.98%, 61.66%, 70.63%, 81.90% respectively, and The dry matters were 20.90%, 32.67%, 43.79%, 57.52%, 66.36%, 74.51%, 82.15% respectively. The results showed that gluten protein is the vegetable protein of low degradation rate.Part â…¡: Effects of gluten protein on movement of the rumen , ruminal fermentation and blood indexes in goatsFour goats fixed with rumen fistula were used to observe the effect of glutenprotein on the rumen movement, metabolism and blood indexes, the results showed that, the experiment period compared with control period, the average contract power of the goat's rumen significantly increased by 31.2% (P<0.05) .The average of goat rumen peristalsis duration time significantly increased by 34.1% (P<0.05) in 15 minutes, also, rumen peristalsis frequency of goats significantly decreased by 25.1% (P<0.05 ) . pH was decreased by 0.64%(P>0.05) ;NH3-N concentration in rumen was increased by 63.87% (P<0.01) ; MCP was increased by 35.18% (P<0.01) ; TDHA was increased by 1.697%,(P>0.05) ;activity of protease was increased by20.72% (P<0.05); in blood , the concentrations of urea nitrogen was increased by 33.70% (P<0.05) ;GH was increased by 8.75% (P>0.05) ;Insulin was increased by 23.09% (P<0.05); T3 was increased by 28.41% (P<0.05) ;T4 was increased by 0.98% (P>0.05) .It suggests that The determine device of rumen movement in this paper can sensitively study law of rumen movement in goats, a certain amount of gluten protein can promote the ruminal fermentation, incense the rumen microbes growth, and enhance the level of concerned hormones.Part â…¢: Studies of rumen fluid with opioid activity from gluten protein by the degradation of rumen microbes on the Xueshan chickensDifferent levels of rumen fluid were added to Xueshan chicken basal diet toincentive growth of chickens by enhanced metabolism of endogenous hormones, the results showed : treat land treat2 ,Compared with control group, food intake increased by4.58% (P>0.05), 5.21%(P<0.05)respectively; body gain increased by 12.09% (P<0.05), 14.84% (P<0.05) respectively; feed/gain decreased by 2.24% (P>0.05) and 3.12% (P>0.05) respectively; in serum, the level of growth hormone were increased by 3.56% (p>0.05), 6.49% (P>0.05) ; the level of triiodothyronine(T3) were increased by 1.36% (P>0.05), 1.05% (P>0.05) respectively; the level of thyroxine(T4) were increased by 19.32% (P<0.01), 26.52% (P<0.01) respectively; the level of insulin were increased by 10.91% (P<0.05), 7.36% (P<0.05) ; the level of Glucagon were increased by 3.01% (P>0.05), 6.79% (P>0.05) . the Newcastle Diease (ND)-Hemagglutination Inhibition (HI) titer of experiment one were significantly higher than the control group in 70 days (P<0.05)... |
