Studies On Immunoaffinity Chromatography (IAC) For Pesticides Multiresidue Analysis

With the prepared antibodies of anti-carbofuran, anti-triazophos and anti-chlorsulfuron, the high, capacity IAC column had been prepared to develop the immunoaffinity chromatography (IAC) technology of carbofuran, triazophos and chlorsulfuron. Based on these, the technology of multi-class immunoaffinity chromatography (MIAC) was developed, the conditions of IAC and MIAC were optimized.0.02mol/L pH7.2 PB was selected as equilibrium and adsorbent medium for carbofuran IAC column and triazophos IAC column, methanol-water (60:40,v/v) as eluent. Under the optimized conditions, the dynamic column capacity was up to1.58μg/mL bed volume for carbofuran IAC column and 1.91μg/mL bed volumn for triazophos IAC column. When the initial concentration of carbofuran or triazophos in standard solution was lower than 2ng/mL, the enrichment efficiency of IAC was more than 167 times for carbofuran or more than 213 times for triazophos. The extract of spiked rice was separated and enriched by IAC, the eluate was detected by antibody-coated direct competitive enzyme linked immunosorbent assay (ELISA) and verified by high performance liquid chromatography (HPLC), every experiment was repeated for 5 times. The results showed that the average recovery of carbofuran from rice was 85.87% with the relative standard deviation (RSD) of 4.71% at the spiked level of 20ng/g. The average recovery of triazophos from rice was 97.93% with RSD of 8.76% by ELISA and the average recovery was 102.5% with RSD of 4.44% by HPLC at the spiked level of 0.1μg/g. The average recovery of carbofuran from river water was 89.75% with RSD of 4.78% by ELISA and the average recovery of carbofuran was 93.13% with RSD of 5.99% by HPLC at the spiked level of 0.1μg/mL .The results from ELISA correlated well with those from HPLC.The chlorsulfuron IAC column, the carbofuran IAC column and the triazophos IAC column were connected in series to separate and enrich chlorsulfuron, carbofuran and triazophos contained in river water and extract of soil, the target compounds in eluate were detected by ELISA respectively, every experiment was reapted for 5 times. The average recovery of carbofuran was 102.4% with RSD of 8.27%, the average recovery of triazophos was 95.01% with RSD of 7.47% and the average recovery of chlorsulfuron was 95.71% with RSD of 9.74% at the spiked levels of 20ng carbofuran, 20ng triazophos and 20ng chlorsulfuron/mL in river water. The average recovery of carbofuran was 92.50% with RSD of 9.02%, the average recovery of triazophos was 94.78% with RSD of 8.96% and the average recovery of chlorsulfuron was 94.39% with RSD of 10.18% at the spiked levels of 100ng carbofuran, 100ng triazophos and 100ng chlorsulfuron/mL in river water. The average recovery of carbofuran was 94.63% with RSD of 8.13%, the average recovery of triazophos was 98.49% with RSD of 11.26% and the average recovery of chlorsulfuron was 98.78% with RSD of 7.84% at the spiked levels of 20ng carbofuran, 20ng triazophos and 20ng chlorsulfuron/g in soil. The average recovery of carbofuran was 97.71% with RSD of 4.65%, the averagerecovery of triazophos was 89.67% with RSD of 6.50% and the average recovery of chlorsulfuron was 92.93% with RSD of 8.46% at the spiked levels of 20ng carbofuran, 20ng triazophos and 20ng chlorsulfuron/g in soil. The results satisfied with the requirement of the pesticide residue analysis.The mixed immunosorbents of carbofuran, triazophos and chlorsulfuron were used to prepare multi-class imrnunoaffinity chromatography (MIAC) column. The 0.02mol/L pH7.2 PB was selected as equilibrium and adsorbent medium of MIAC column and methanol-water (80:20,v/v) as eluent. Under the optimized conditions, MIAC was applied to clean up and enrich carbofuran, triazophos and chlorsulfuron from river water and extract of soil, the target compounds in the eluate from the MIAC were detected by ELISA and verified by HPLC, every expriment was reapeated for 5 times.
