In long domestication history, silkworm Bombyx mori has formed different geographical populations or ecological type. There are several varieties in each population, its ecology and economic properties each have characteristics. Study on hereditary difference, classification, evolution and relationship between different geographical varieties, not only the basis of determination, keeping and utilization of silkworm germ plasm, but also the foundation of variety cultivating and popularization. So it has been paid close attention for a long time. Silkworm Bombyx mori originates from Bombyx mandarina is generally acknowledged for a lot of scholar, but there are many questions we couldn't resolve, e.g. it how originated and Bombyx mori system how divided, etc. In addition, the studies on silkworm origin and system division developed slowly in recent years, maybe, the new molecular marker could provide some new important information and idea.In present study, 7 silkworm Bombyx mori and B. mandarina Ankang were investigated with the fluorescent semi-automated detection technology (GeneScan) , 11 microsatellite loci were used. We want to provide some molecular theoretical foundation for molecular evolution of silkworm, besides we discussed the sampling strategy. Some results of the study is as follows:1. Comparison of the RAPD, AFLP and SSR: With the same materials, RAPD, AFLP and SSR were done. The important genetic parameters were analyzed and compared, the results showed that SSR technology has high polymorphism information content (PIC), relatively suited to the study of molecular evolution.2. Among the 11 microsatellite primers, amplification of 9 pairs is good. The 9 microsatellite primers detected 108 alleles. Each primer detected range 5 to 17 alleles. The average of each primer, detected alleles, was 12. The proportion of polymorphic alleles was 100%. The polymorphism information content was between 0.2437-0.5603.
