Botanic sterilants are made up of the extracts of natural plants which have the effect of anti-fertility. Botanic sterilants have characteristics of instant effect ,high efficacy, perfect eidbility to rodent , wide resources,low costs,no pollution,no posioning to mouser, no manslaughtering of benefical animal,no drug-resistance in constant use,and lead to no damage to crops with a few of rodents.Such bait made from botanic sterilants can seriously damage rodent's reproductive organ,block the reproductive process, reduce population .In order to investigate and understand the anti-fertility mechanism of gossypol and triperygium wiffordii further, the histological,ultrastructure and endocrine effects of these two kinds of botanic sterilans were investigated in mandarin vole(Microtus mandarinus) first time. Histological method was used to study postembryo development of the testis and epididymis' and effect of gossypol and tripergium wilfordii on mandarin voles(Microtus mandarinus ).Immunocytochemical method was used in the research for locating distribution of testosterone, estradiol and their receptor on the process of spermatogenesis. This method was also used to study the effect of gossypol and triperyguim wilfordii' on the expression of testosterone and estradiol and their receptor in testis and epididymis.The immunocytochemical method was also used to probe into the postembryo's development of mandarin voles's testis and epididymis,the law of spermatogenesis and the anti-fertility mechanism of gossypol and triperygium wifordii. 1.The histological development of mandarin vole's testis and epididynis:Sort out 6 groups of mandarin voles,each including 6 ones,aging from 1day(24hours),10 days,25 days,45 days,60 days to 70 days,The method of microstructure and ultrastructure were used to show postnatal development process of the testis of mandarin voles. Results show that seminferous tuble of diameter as well as germinal epithelium's cell develops as fast as the increase of age. Germinal cells are mainly goncytes for one-day mandarin voles, spermatogenous cells increase and divide obviously for ten-day's, spermatids are generated for twenty-five day's, matual sperms come into being for forty-five day's,all kinds of germinal cells emerge and lots of matual sperms in seminferous tuble and epididymis were produced forsixty-day's.It's concluded that mandarin voles reach the age of puberty on the 45th day,mature at about 60 th day generally keep the same pace with the development of testis.2. Function of androgen and esterogen as well as their receptor to the postnatal development of the testis and epididymis : While the testis and epididymis' structural research is going on,observation of androgen and estrogen as well as their receptor 's expression on testis and epididymis is also observed using method of immunocytochemical . Testosterone and AR's effect is obvious in their process of development, especially in 25-45 th day, while the diameter of the seminiferous tubule became wider as well as germinal epithemium became thicker .The result may infer that combining testosterone with AR could promote and preserve the development of semiuiferous tubule and hyperplasia of germinal epithelium. With the interposition of ER α estrogen can restrain seminiferous cells form apoptosis,promote the development of epididymis. T and AR express at different stages of epididymis development relying on testosterone.3.The function of testosterone and AR in process of spermatogenesis: the combination of Androgen and androgen receptor adjusts spermatogenesis . Immunocytochemical results showed that testosterone expressed mainly in the spermatid of mandarin voles.'testis development and strongest in the sperm of adults which are superior to spermatocyte and spermatid. AR present spermatid in its development, spermatocytes and sperms express much stronger in adult .The result mat infer that combining testosterone with AR preserved spermatogonium vitality and hyperplasiability promote spermatocyte meiosis, participate in spermatid anomalism and preserve mature sperm energe. Leydig's cell produce testosterone and adjust it's synthsis function by means of autocrine mode .Leydig's cell can also regulate the secretion of Sertolis cell by means of paracrine to control sperm creation. Expression of T and AR in epididymis suggested that the function of epididymis is regulated by testosterone .4. The function of estradiol and ER α in the process of spermatogenesis: The combination of estradiol and ER α located in sperms bring regulation into play,Immunocytachemical result showe that estrogen expresed effectiverly in spermatocyte and spermatid. ER α expreses obviously in spermatocyte ,but not better than in sperm and spermatid.From all above ,we can infer that estrogen had a...
