| As one of the most important economic crop and extensive cultivated vegetables, Chinese cabbage (Brassica campestris L. ssp. Pekinensis) were originated from China. It is the main exported vegetable. But the tipburn in Chinese cabbage has a great affect on the exportation. It is a physiological disorder induced by calcium deficiency. Different variety has significantly difference in resistance to tipburn. Therefore, breeding the resistant variety to tipburn disorder is the hotspot in Chinese cabbage breeding. A population of F2 generation including 115 individual plants was derived from the cross between the resistance lines F1-6-5-2-2-2-3 and the susceptible lines P14-2-1-15.In this paper, the AFLP markers were employed in constructing a molecular genetic map and mapping QTL controlling tipburn disorder resistance in Chinese cabbage. The main results were as follows: 1. Through the comprehensive selection in many lines by the detached leaves inserting into detection solution method and combination with the phenotypic trait in field observed by the Qsingtao Agricultural Science Research Academy, we acquired the resistant line F1-6-5-2-2-2-3 and the susceptible line P14-2-1-15.And the F2 population were derived from the parents. 2. Thirty-two pair of optimum primes were selected from 192 pair of primes by parents.E01/M02 prime combination was used in pre-amplification and E+3/M+3 prime combination was used in amplification. 3. A molecular genetic map of Chinese cabbage was constructed using AFLP markers by Joinmap3.0 software. 105 markers were integrated... |
