In the work , we studied effects of low temperature on nematode activities and designed the equipment of cold brake in terms of it. By means of inoculating pine wood nematode, axenic pine wood nematode , GcM5-1A,axenic pine wood nematode mixed with GcM5-1A and the secretion of GcM5-l A and AM2B we further studied pathology of BX. In addition, the effects of aloperine on pine wood nematode is studied. The main results are as follows:1.we studied effects of low temperature on pine wood nematode activities and achieved a limit of low temperature which pine wood nematode could not get over in terms of it we designed equipment of cold brake ,and we located BX in local positions of pines by means of physical methods ,so that they could not move to other bodies of pines .2. Axenic pine wood nematode , GcM5-lA and Axenic pine wood nematode mixed with GcM5-1A were respectively inoculated on three-year-old pine . 60 days later ,the pine trees which were only inoculated Axenic BX and GcM5-1A still grow healthy, but pine trees which were inoculated Axenic BX with GcM5-1A died . So we can see that only Axenic pine wood nematode or GcM5-1A could not induce the trees die ,only Axenic pine wood nematode with GcM5-1A could . The outcomes of examinations validated that the pin wilt disease was a complex-disease which arosed by pine wood nematode and pathogenic bacterium3. The pine trees which were inoculated the toxin of Pseudomonas fluroescens GcM5-1A died , but the trees which inoculated the secretion of P.asaccharolyticus AM2B not died .It showed that the pathogenic bacterium that BX take could produce toxin which could lead the trees die ,the without-pathogenic bacterium could not lead tree died .Because of the same conditions it also settled the doubt that the toxin which could make the trees die did not produce from culture medium or any other approaches .4.we studied the effects of aloperine on activities of pin wood nematode by two-layer culture medium .It showed that aloperinec could drastically restricted pine wood nematode movements , which also explained why the mixture of aloperinec and antibiotics could increased the killing of pine wood nematode, and it provided evidence for Lysenkoism of complex-disease and selection of nematicides。 The innovations of the dissertation :1. After studying the effects of low temperature on pine wood nematode activities , we designed equipment of cold valve which could located pine wood nematode in local positions of pines.2. A series of Laboratory inoculation by cold valve directly proved that the pin wilt disease is a complex-disease involving pin wood nematodes and pathogenic bacterium.3.There is doubt that the pathogenic ability of pine wood nematodes influenced by themotility .The study indicated that there is no inevitable relationship between the pathogenic ability and motility .The pathogenic ability maybe correlate to the kinds of pathogenic bacterium that the pine wood nematode take .4. Cold valve is a new method which is suitable for inoculations to adult pine and in the open door outside asepsis , it can control the movement of eelworm and exclude the disturbance of animalcule, it also bring forth a new way for settling other complex-disease .
