Soybean seeds contain three lipoxygenase isozymes designated L-l,L-2,L-3 that can account for approximately 1% of total seed protein. Lipoxygenase which catalyzes,as a primary reaction, certain unsaturated fatty acids occur oxidation,and the action generates the hydroperoxidation. Lipid hydroperoxides,which are formed during oxidation,can directly bind protein and other nutritious compounds and also led to the destruction of certain vitamin and protein,and their breakdown products can have toxic effects.The oxidation products further decompose to middle chain aldehydes and alcohols et al which are major constituents of the undesirable grassy-beany flavor and bitter taste in soybean products. Soybean lipoxigenase is the most limiting factor to control soybean nutrient value and flour. As following, People in science research have studied to reduce the problem during food processing of soybean by varieous techniques.However,these treatments are expensive and not entirely satisfactory for food materials because the soybean proteins have poor solubility caused by heat treatment.Through making advantage of shortcut and effeciency assays,we can selectively detect lipoxygenase lacking mutants,and use soybean cultivars as acceptors and cross with iso-genic mutants as donors for this character,which is an available approach.In a word,it is urgent to designate a simple,sensitive,and rapid mean of determining lipoxygenase in soybean seeds.Based on citing articles,we have known that soybean lipoxygenase is 94 to 97KD enzyme with water-solubility,which is larger than 60 000 dal as complete antigen according with immunoassays.MonocIone antibodies(MAbs) of lipoxygenase-1(Lox1) were produced and detection methods were established using the MAbs.Isoelectric focusing in thin-layer polyacrylamide gels(IEF-PAGE) has been applied to the analysis of the enzymes in near iosogenic line(NIL) of soybeans, For examplelipoxygenase null-2,lipoxygenase-l band is moved to positive;given...
