Insect pest is one of disasters to crops in agriculture production. In the past, chemical insecticides were used to control the pests, which often caused the pollution and contamination to environment. Transgenic plant assistant to pest were obtained by using biotechnology. Both scorpion neurotoxin gene and chitinase gene are valuable for producing transgenic crops. Tobacco (Nicotiana tabacum L) is widely used species in life science as a mold plant because it is easily operated in plant culture and strong ability of regeneration. Study on the co-expression of extrinsic gene in transgenic plants using the mold plant tobacco was significant in investigation of expression condition of the anti-insect gene.In this paper, binary recombinant plasmids pBI101-Chi-BmKITS containing scorpion toxin BmK ITS gene and Chitinase gene were used to generated two plasmids pBI101-BmKITS and pBI101-Chi, which contain BmKITS gene and Chitinase gene, respectively. Two plasmids were used transformed tumefacience LBA4404(St~r) to obtain the engineering Agrobacterium strains harboring the target genes. All strains were tested by using PCR procedure.Transformation of tobacco mediated by A. tumefacience were performed by using the procedure of leaf-dish. Some valuable data have been obtained. It would provide basal data for next work.
