In this paper, different explants (segments, leaf, anther) tissue culture of Caragana microphylla Lam were studied systematically. The results were as follows:MS was the optimal medium, which was used for callus induction of anthers, leaves and propagation of segments.Twig was the optimal explant in the cultivating of segments, tender leaves and segments were the optimal explant for inducing callus, In the cultivating of anther, the bud of middle uninucleate and late uninucleate were the optimal explant.The different hormone concentration with 6BA and 1AA was beneficial to segments multiplication and rhizogenesis. The different hormone concentration with 6BA and 2,4-D was beneficial to callus induction. The results showed that the optimum medium of inducing bud was MS+6-BA0.5mg/L+ IAA0.01mg/L. The optimum medium of rooting was 1/2MS+IAA0.5 mg/L.Sucrose played a role in the callus induction within a certain concentration, but high concentration will restrict callus induction. In the culture of segments, 3% sucrose was the optimal concentration. In the culture of anther, 6% sucrose was the optimal concentration.Low temperature pretreatment with 4℃ for 3d on buds before inoculating was beneficial to callus induction;Heat shock with 30℃ for 1d after inoculating was also contributed to callus induction.
