Salt stress is one of the major abiotic stress in agriculture worldwide. In the study, two dimensional electrophoresis (2-DE) and MS was used to analyse the proteome of the salt-tolerant mutant of wheat (RH8706-49) treated with 150mM NaCl for 1,6,12and72h. Total proteins of roots were extracted and separated by two-dimensional gel electrophoresis.there are 16 spots were reproducibly detected, including 9 that were up-regulated and 7 down-regulated. After 3 candidate proteins are identified expressed differently in gels being analysised by MALDI-TOF-MS and Mascot software, the there candidate proteins were identified: H+-transporting two-sector ATPase, F1-ATPase alpha subunit and putative dihydrolipoamide dehydrogenase. Using the same protocol, the root proteome is compared between the salt-tolerant mutant of wheat (RH8706-49) and the salt-sensitive mutant of wheat (H8706-34) treated with 150mM NaCl for 48h.. There are 6 candidate protein spots are selected for MS.We postulated the proteins unmatched to known proteins in the database were new proteins.The six spot were identified three peptides by MS/MS. A cDNA fragment which include peptide is obtained by RT-PCR. Implications of these proteins for the molecular mechanism involved in salt-tlerance are discussed.
