Salinomycin sodium is one of the polyether ionophore antibiotics drugs in animals and the most used feeding additives and very effective in promoting growth and anti-coccidiosis on pigs and fowls.But poisoning incidents on pigs and fowls reported can be seen in many messages. But the reports on the study of toxicity machanism is difficult to see inner and outer China. The experiment included acute toxicity and subchronic toxicity. Acute toxicity was conducted to determine the median lethal dose (LD50) of salinomycin sodium to rabbits and seeked the most effective level in rabbit feeds. This experiment mainly studied the physiological and biochemical indexes, and pathological changes of livers, lungs, kidneys, et al. The aim of this experiment was to elucidate the toxic mechanism of Salinomycin sodium on drabbits.Advanced test. Fifteen healthy rabbits between 0.75 1.0kg were chosen. Salinomycin sodium was orally administered to them.Two weeks later ,the 100% and 0% lethal doses were gained. The dose was 150 and 50mg/kg body weight. The geometric proportion series was 1.3 according to simpled Karber way which decide the acute toxicity test doses were 50,65,85,111,144mg/kg body weight.Acute toxicity test. Thirty rabbits between 0.75 1.0kg were randomly allotted five groups, six rabbits each group. Salinomycin sodium was orally administered to these groups through 144 to 50 mg/kg body weight respectively. When the dose went beyond 144mg/kg, that group appeared 100% dead. Estimated by simpled Karber way, the LD50 was 91.2 mg/kg body weight, which ninety-five per cent confidence interval is 106.86 77.84 mg/kg body weight and standard error is 0.0351. The onset of clinical signs varied from 1 hour to 6 days after administration of the drug, and clinical courses lasted from 2 to 6 hours. Clinical signs included tachycardia and cardiac arythmia ,incoordination sternal or lateral recumbency, paddling and death. Both gross and microscopic lesions were observed in livers,lungs and kidneys.Subchronic toxicity test. Fifty-two healthy rabbits were chosen for this part. They were randomized equally on a stratified body weight basis into four groups with a control and three trial groups. The four formulated diet contained Salinomycin sodium 0 mg/kg( I ), 100 mg/kg (â…¡) ,75 mg/kg (â…¢) ,50 mg/kg (â…£) mg/kg respectively according to LD50. It was including earlier period (120d ) and later period (2040d). During the trial the clinical manifestations were observed whenever, weighed one by one and recorded feed consumption weekly. At the end of each feeding period, drew randomly five rabbits out of every group, stripping hearts, blood samples were collected in heparinized tubes for haematological analysis included the content of hemoglobin ,the number of red blood cell and number of classified white blood.
