The licorice (Glycyrrhiza uralensis fisch), whose roots and rhizomes are most common Chinese herbal medicine, is a kind of perennial suffurticosa herbal plants from Glycyrrhiza species Legume family. Besides medical value, the licorice is widely used in food, beverage, cigarette, cosmetics and other industries as additive. As natural resources can't fulfill require of medicine and other industries. Cultivation and tissue culture are considered to be hopeful way to alleviate the crisis of natural resources. Tissue culture technic is a good method, it can control grow condition of plants, accelerate pace of propagate, and distill effectual element from cells. Glycyrrhiza uralensis Fisch as material in the test. But for licorice tissue culture technology was not mature only few papers were reported, there are no reports about Somatic embryogenesis and effect of hormone to callus. We study the course of callus, effect of hormone to callus, suspension culture, observe the course of Somatic embryo through espial of cytology, intent to set up system of induce callus. Defined the callus' state to translate Somatic embryo, offer theory and technic foundwork of propagation and distill effectual element from cells, The rusult as follow:1. Explent began to appear callus after 3 days, the callus occur from morphologyic extremity, extend to the whole expelnt along axis.2. Study the effect of hormone and their collaboration to induce callus. Callus can't be induced on MS culture medium with single hormone, the best combination of induce callus is 0.2mg/L NAA +1.0mg/L BA +1.0mg/L 2,4-D.3. Study the effect of hormone and their combination to callus continue culture, when the callus was cultured on MS culture medium with 6-BA 1.02.0mg/Land lower concentration of ZT, KT, then embryogenic callus appeared. We obtained callus to suspension culture and brown and green, granule embryogenic callus.4. Study the effect of suspension culture through hormone and additional matter, establish tissue culture's suspension system: MS culture medium with 0.2 mg/L NAA +0.1 mg/L BA +0.1 mg/L KT. Get Somatic embryo in the suspension system.5. In suspension culture, cell grow curve show "S" , The time of logarifhim growth appears after inoculation 10-20 days; Mass wastage of sucrose in logarithim growth; Mass wastage of NH4+ in front and behind logarithim growth, pH and Electric conductivity' tide like NH4+, they are indicate as nutrition wastage.
