A somatic embryo regeneration system of Chinese early-fruiting walnut(Juglans regia L.) and a epicotyl regeneration system are acquired through adjusting the composition and the concentration of plant growth substances in this research.The somatic embryo regeneration system on Chinese early-fruiting walnut is the first reported and will produce enough materials for the latter drought gene transformation. Conclusion are as follows:1. 6-BA sal medium of DKW and culture conditionThe pH of DKW 6-BA sal medium between 5.8 and 6.2 is better for induction of embryonal callus and usage of organic nitrogen can not inhance the effect of induction of embryonal callus significantly. The highest induction is acquired in the concentration of 55 g/L and 45 g/L cane sugar.2. Difference of induction of different walnut explantsyuanfeng cotyledons induction of embryonal callus is 16.5% in the medium of DKW with KT 2mg/L+6-BA 1mg/L+IBA 0.01mg/L.There is no significant difference among somatic embryo inductions of three cultivar walnut immature embryos after 67-97 days of pollination. Somatic embryo induction of hypocotyl is 62.60%.3. Induction of somatic embryo and secondary somatic embryoThere is no significant difference in the three plant growth regulator combination. The somatic embryo induction of the combination KT 2mg/L+6-BA 1mg/L+IBA 0.01mg/L is the highest,which is 68.00%. Different mediums have significant induction of somatic embryo and the best one is 42.88% in the condition of DW medium with 55g/L cane sugar and 250 mg/L glutamine.
